Specifications include, but are not limited to:The project entails building an electronic reader for performing remote diagnostic assays in austere environments, as part of a project funded by the Bill and Melinda Gates Foundation. The reader heats cartridges and performs fluorescence-based assay measurements, then sends results to a smartphone or tablet through an attached cable. The device needs to be rugged, low-cost, operable in daylight and in environments in which electricity is not available, and should provide accurate, reproducible measurements of the fluorescent output produced by diagnostic assays. The project will ideally entail two phases. In Phase 1, the vendor will perform reader development to create a bench-top proof-of-principle instrument that meets the required technical criteria. The vendor will perform fluorescence calibration tests on this prototype to demonstrate feasibility. If successful, in Phase 2 a second development/build cycle will create a portable reader prototype that meets the performance criteria demonstrated in Phase 1 and incorporates the design and environmental constraints for our end-users. The vendor will deliver two completed readers at the end of the project that include the following components: 1) smartphone/tablet app that quantifies fluorescence measurement data from the reader and presents the results to the user; 2) optimized portable reader; and 3) product demonstration. During the course of the project, the developer will work closely with the customer and provide weekly or bi-weekly progress through meetings for planning purposes. SPECIFICATIONS & INSTRUCTIONS FOR COMPLETING SPECIFICATION TABLE: The following specifications represent minimum acceptable requirement. Bidders must select either yes or no for each specification below. For any No Response, the bidder must provide a detailed written explanation indicating how this difference in specification would affect the performance of the product being bid. The University shall be the sole judge of acceptability. MEET SPECIFICATIONS ITEM DESCRIPTION YES NO 1 Reader development will be provided through a 2-stage feasibility study; in Phase 1 the optical path/processor and heater/thermocouple will be developed to demonstrate sensitivity to our target fluorescence probe concentration (defined below); in Phase 2 the optimized optical path and heating system will be engineered into a hand-held prototype. 2 The final prototype must have the approximate dimensions of a can of Red Bull (~7 inches tall; ~3 inches wide). A cylindrical shape with a flat base is ideal. The minimum product dimensions are 3 wide and 4 tall and maximum product dimensions are 4 wide and 8 tall. 3 Reader will heat a transparent polymer assay cartridge (sample; ~2-2.5 cm wide or on an edge) at a defined temperature within the range of 40-55 C and hold the sample at a constant temperature (+/- 1 degree) for 15 min. 4 Reader heater must be optimized to use minimal power and must be able draw current from an on-board rechargeable battery and the smartphone/tablet. 5 Imaging hardware must consist of off-the-shelf controllers, boards, LEDs, cameras that will meet the desired assay sensitivity (defined below). 6 The imaging system must have sufficient sensitivity to detect an aqueous solution of 50 nM of 6-carboxyfluorescein in a 500 micron-tall chamber in the transparent polymer assay cartridge. Customer will provide reagents and cartridges. 7 The reader must be able to perform >5 assay measurements before it requires recharging. Recharging is ideally performed rapidly (<6 hr). The measurements can be fixed time measurements such that the light source and camera only require being turned on 1-2 times per cartridge.
